Morphology and Replication of Intranucleolar Dna in Polytene Nuclei
نویسنده
چکیده
This report is concerned with filamentous intranucleolar structures in the polytene salivary gland cells of Drosophila melanogaster . A number of studies on a variety of cells have provided evidence for the chromosomal association of such structures (e .g. references 6, 7), fortifying the concept of Lettre et al . (11) that the nucleolonema, of historic controversy, is not an autonomous structure of the nucleolus, but instead is derived from a portion of the nucleolar chromosome . Support for that concept and, further, a basis for the proposition that all of the filamentous structures of the nucleolus have a common identity, may be found in this study of a single cell type in which intranucleolar structures of diverse morphological conformations ranging from a dense mass to a much extended, ramified filament have been observed . A series of micrographs are presented as evidence that all forms are homologous and are alternate forms of the laterally extruded nucleolus organizer locus of the chromosome complement . Radioautography of thymidine incorporation confirms that these structures contain replicating DNA and indicates that a negative correlation exists : the dynamic morphology is not related to replicative activity .

Replication in Drosophila chromosomes. I. Replication of intranucleolar DNA in polytene cells of D. nasuta.
The organization and replication of intranucleolar DNA in polytene cells of Drosophila nasuta have been examined. Normal Giemsa and fluorescent (Hoechst 33258 and quinacrine mustard) staining reveal that in D. nasuta polytene cells, the intranucleolar DNA is organized into condensed and disperse forms and very often shows connexion to the alpha heterochromatin of the chromocentre. Results of [3...

We have used Xenopus laevis cell-free extracts to study patterns of DNA replication in polytene nuclei isolated from salivary glands of Drosophila melanogaster 3rd instar larvae. Replication was visualized by supplementation with biotin-dUTP so that nascent DNA became labelled, thus allowing detection with fluorescein or Texas-Red-conjugated streptavidin. Biotin incorporation was dependent on i...
متن کاملTemperature-dependent functional structures in the polyteene chromosomes of Phaseolus, with special reference to the nucleolus organizers.
During the endopolyploidization of the suspensor cells of Phaseolus coccineus and P. vulgaris, polytene chromosomes and an important nucleolar apparatus develop within the nuclei. The nucleolus-associated regions of the four nucleolus-organizing chromosomes and the regions of the secondary constrictions (intranucleolar chromatin threads) show a temperature-dependent, reversible variability in t...
متن کاملDistribution of DNA in heterochromatin and euchromatin of polytene nuclei of Drosophila hydei.
HE concept that all cells of an organism contain the same quantity of DNA Tor a geometric multiple of it has been demonstrated in a large number of cases (SWIFT 1950; MIRSKY and RIS 195 1 ) . Many exceptions to this concept have been observed, some recent ones being those described by BEERMANN ( 1959,1966) and GEYER-DUSZYNSKA (1 966). For some organisms, it was demonstrated that the exception t...
متن کاملFine Structural Alterations of Interphase Nuclei of Lymphocytes Stimulated to Growth Activity in Vitro
This report describes fine structural changes of interphase nuclei of human peripheral blood lymphocytes stimulated to growth by short-term culture with phytohemagglutinin. Chromatin is found highly labile, its changes accompanying the sequential increases of RNA and DNA synthesis which are known to occur in lymphocyte cultures. In "resting" lymphocytes, abundant condensed chromatin appears as ...
